Diseases caused by trypanosomatids are endemic in certain geographical areas and represent a serious public health, economic and political problem, as their annual incidence is increasing, particularly in poor or developing countries. Among the diseases caused by these parasites, leishmaniases and Chagas disease predominate in South America. Patients afflicted with these diseases do not receive financial support from the government or from industry, and the drugs used to treat affected patients in the clinic are ineffective and highly toxic. Moreover, there are no vaccines for these diseases.
The chapters in this book were developed by a multidisciplinary group of researchers with substantial experience studying different aspects of these diseases, including the processes associated with developing new compounds with anti-trypanosomatid activity. These chapters focus on the immune responses and inflammatory reactions elicited by various compounds under testing, as well as the biochemical and cellular and molecular biological mechanisms underlying these responses and reactions. These chapters also discuss the best animal models for performing specific in vivo studies of the activity and toxicity of promising compounds that may eventually be used for the treatment of the above mentioned pathologies, as well as the effects elicited by specific drug combinations and the problems caused by parasite resistance to different compounds.
Given the above mentioned difficulties associated with treating leishmaniases and Chagas disease, it is essential that researchers identify compounds enabling the development of new highly effective drugs without side effects that do not easily induce resistance. Here, we discuss the most important issues associated with such research endeavors, including the chemical and biological aspects of these issues.
